Do you have to register for one of these limited positions to use First Strike rounds at all at this event?

No. But, in order to take part in the specific scenario missions (I've no clue what they are yet), you must apply and be approved for the mission and, use the markers issued by Tiberius/Viper.

Hate to Necro but, just what happened with these missions last year?

I've not seen any recap, articles or even scores related to them. Just the announcement of who would be doing them.
